I'm currently running TFF 24.3 and want to update
Must I do incremental updates or can I go directly to 45.4?
Will I need to reset my profile?
IMPORTANT! Will updating cause loss of my bookmarks; I have several hundred.

This should work without a profile reset, but I'd make the following recommendations:
Export your bookmarks to an HTML backup first. Bookmarks > Show All Bookmarks, and on the window that opens, click the "star" icon in the toolbar and select Export Bookmarks to HTML. (The Import option is in the same place if you need it.)
Although I think you'll be okay skipping these steps, just to be safe, upgrade to 31.8.0, update to 38.10.0 and finally update to 45.4.0.
If the browser behaves strangely when you finally get to 45.4.0, then I'd do a profile reset first thing. Doing so should save your bookmarks, but if it doesn't, you have the HTML backup.
